Systemic reovirus therapy of metastatic cancer in immune-competent mice.

The human reovirus is an oncolytic virus that specifically targets cancer cells with an activated Ras pathway. Because it is replication competent and highly specific for cancer cells, this virus has the potential to be an effective antimetastatic cancer agent through remote site delivery. In this study, we exploited the ability of reovirus to replicate in murine cells to test the efficacy of t...

Oncotargeting by Vesicular Stomatitis Virus (VSV): Advances in Cancer Therapy

Modern oncotherapy approaches are based on inducing controlled apoptosis in tumor cells. Although a number of apoptosis-induction approaches are available, site-specific delivery of therapeutic agents still remain the biggest hurdle in achieving the desired cancer treatment benefit. Additionally, systemic treatment-induced toxicity remains a major limiting factor in chemotherapy. To specificall...

Inhibition of a vesicular stomatitis virus mutant by rifampin.

Rifampin reversibly inhibits the intracellular replication of a VSV mutant, whereas a revertant variant selected from this viral population, as well as the wild strain, are not affected by this drug. Rifampin inhibits transcriptase activity of the sensitive mutant only and, consequently, total viral RNA synthesis decreases significantly in the cells.

Oncolytic vesicular stomatitis virus for treatment of orthotopic hepatocellular carcinoma in immune-competent rats.

Tumor-targeted replicating viruses are being developed as a novel class of oncolytic agents. Vesicular stomatitis virus (VSV) is a negative-strand RNA virus with inherent specificity for replication in tumor cells due to their attenuated antiviral responses. VSV as an oncolytic virus is particularly appealing for its exceptionally rapid replication rate in tumor cells, such that the oncolytic e...
